Role of NF-κB and Upstream Kinases in the Evolution of Pancreatic Carcinoma

Background/State of the art. The transcription factor NF-κB and its upstream regulatory network have been implicated in many different aspects of tumor development and progression. An interesting feature of NF-κB regulation is the presence of two distinct activation pathways, the canonical pathway (involving the IKK subunits IKK2 and NEMO, and p50/RelA dimers), and the non-canonical pathway (involving the kinases NIK (NF-κB-inducing kinase) and IKK1, and p52/RelB dimers). In pancreatic ductal adenocarcinoma (PDAC), the role of the canonical NFκB signaling pathway has been highlighted in mouse models, but there is also evidence for an important role for non-canonical signaling (Huber et al., 2004; Prabhu et al. 2014).
